Introduction: One main limitation in biomarker studies using EVs is the lack of a suitable isolation method rendering high yield and purity samples in a quick and easily standardized procedure. Here we report an affinity isolation method with a membrane-sensing peptide (MSP) derived from bradykinin. Methods: We designed a protocol based on agarose beads carrying cation chelates to specifically bind to the 6His-tagged membrane-sensing peptide. This approach presents several advantages: 1) cation-carrying agaroses are widely used and standardized for His-tagged protein isolation, 2) the affinity protocol can be performed in small volumes, feasible and manageable for clinical routine and 3) elution with imidazole or EDTA allows a gentle and easy recovery without EV damage, facilitating subsequent characterization and functional analyses. Results: The optimized final procedure incubates 0.5 mg of peptide for 10 min with 10 µL of Long-arm Cobalt agarose before an overnight incubation with concentrated cell conditioned medium. EV downstream analyses can be directly performed on the agarose beads adding lysis or nucleic-acid extraction buffers, or gently eluted with imidazole or EDTA, rendering a fully competent EV preparation. Discussion: This new isolation methodology is based on the recognition of general membrane characteristics independent of surface markers. It is thus unbiased and can be used in any species EV sample, even in samples from animal or plant species against which no suitable antibodies exist. Being an affinity method, the sample handling protocol is very simple, less time-consuming, does not require specialized equipment and can be easily introduced in a clinical automated routine. We demonstrated the high purity and yield of the method in comparison with other commercially available kits. This method can also be scale up or down, with the possibility of analyzing very low amounts of sample, and it is compatible with any downstream analyses thanks to the gentle elution procedure.

La residencia en psicología clínica es un proceso que pondrá al límite la capacidad de adaptación de los residentes,especialmente si no gozan de la supervisión y el soporte institucional adecuados. Conscientes de esta situación, estetrabajo pretende normalizar la complejidad que supone formarse como psicólogo clínico en España, poniendo unénfasis especial en los primeros pasos como residente en el Sistema Nacional de Salud y en los desafíos personalesque ello implica. Asimismo, persigue incrementar la sensibilidad sobre la necesidad de medidas que permitan a losresidentes sobrevivir a la psicología clínica. Por ende, se discute el estado del arte sobre el concepto de autocuidadodentro del campo de la psicología clínica y la psicoterapia desde la perspectiva de que especialistas mejor formadospodrán mejorar el Sistema Nacional de Salud y la atención a la salud mental de los más vulnerables.(AU)


The training system in clinical psychology in Spain is a process that will test the adaptability of trainees to thelimit, especially if they do not have adequate supervision and institutional support. Aware of this issue, this paperaims to normalize the complexity of training as a clinical psychologist in Spain, placing special emphasis on thefirst steps as a trainee in the National Health System and on the personal challenges involved. Moreover, it seeksto increase sensitivity and awareness about the importance of establishing measures that enable trainees to surviveclinical psychology. Therefore, the state of the art on the topic of self-care within the field of clinical psychologyand psychotherapy is discussed from the perspective that better trained specialists will improve the National HealthSystem and mental health care for the most vulnerable people.(AU)

La terapia de aceptación y compromiso (ACT) pone de manifiesto la importancia de la evitación experiencial y la fusión cognitiva en un amplio espectro de problemas psicológicos. El propósito de este estudio es conocer cómo influyen la gravedad del trastorno de pánico y agorafobia (TPA) y la gravedad del trastorno obsesivo-compulsivo (TOC) en los niveles de evitación experiencial y fusión cognitiva. Se pretende examinar, además, si existen diferencias entre ambos grupos diagnósticos. La muestra estaba constituida por 67 personas diagnosticadas de TPA y TOC. Se utilizaron los siguientes instrumentos de evaluación: Acceptance and Action Questionnaire (AAQ), Cognitive Fusion Questionnaire (CFQ), Yale-Brown Obsessive Compulsive Scale (Y-BOCS) y Beck Anxiety Inventory (BAI). Los resultados mostraron que los pacientes con TPA presentan mayores niveles de fusión cognitiva frente a los pacientes con TOC, que elevados niveles de ansiedad se asocian a mayor grado de evitación experiencial y que una mayor gravedad del TOC correlacionaba con elevados niveles de evitación experiencial y fusión cognitiva. Se propone una intervención temprana en pacientes con TOC para prevenir la cronicidad del trastorno y se recalca la importancia de los aspectos de aceptación en el trascurso de la terapia de estos pacientes (AU)


Acceptance and Commitment Therapy (ACT) highlights the importance of experiential avoidance and cognitive fusion in a wide spectrum of psychological problems. The purpose of this study is to know how the severity of panic disorder and agoraphobia (APT) and the severity of obsessive-compulsive disorder (OCD) influence levels of experiential avoidance and cognitive fusion. It is also intended to examine whether there are differences between the two diagnostic groups. The sample consisted of 67 people diagnosed with APT and OCD. The following assessment instruments were used: Acceptance and Action Questionnaire (AAQ), Cognitive Fusion Questionnaire (CFQ), Yale-Brown Obsessive Compulsive Scale (Y-BOCS) and Beck Anxiety Inventory (BAI). The results showed that patients with APT present higher levels of cognitive fusion compared to patients with OCD, that high levels of anxiety are associated with a higher degree of experiential avoidance, and that greater severity of OCD correlated with high levels of experiential avoidance and fusion cognitive. Early intervention in OCD patients is proposed to prevent the chronicity of the disorder and the importance of the acceptance aspects is emphasized in the course of therapy of these patients (AU)

BACKGROUND: Knowing the feed intake pattern during lactation of modern genetic sows is crucial because it allows to anticipate possible problems and maximize their performance. On the other side, electronic feeders permit real-time data to be available for a more accurate evaluation of sow eating behavior. This work aimed to characterize the feed intake patterns of lactating highly prolific sows and determine their effect on reproductive performance. A database of 1,058 registers of feed intake collected from a commercial farm was used to identify five consistent sets of clusters (feeding curves) using machine learning. In the second step, the five feeding curves were characterized into five patterns by high, medium and low feed intake during 0-6 d and 7-28 d of lactation: 1-HH, 2-MH, 3-HM, 4-MM and 5-LL. RESULTS: The mean daily feed intake of all the sows was 6.2 kg (0.06 SEM) across the 5 patterns. As the pattern numbers increased from 1-HH, 2-MH, 3-HM and 4-MM to 5-LL, their mean daily feed intake decreased from 7.6 to 6.9, 6.4, 5.8 and 4.3 (0.06 SEM) kg, respectively (P < 0.01). Sows with Pattern 1-HH tended to have shorter weaning-to-first service interval (P = 0.06) and had a higher farrowing rate than those with Pattern 5-LL (P < 0.01). Furthermore, contrast analysis showed that sows with Patterns 1-HH and 2-MH tended to have more piglets weaned (P = 0.05) and lower preweaning mortality (P = 0.07) than those with Patterns 3-HM and 4-MM. Also, sows with Patterns 1-HH and 3-HM had fewer stillborn piglets and a lower percentage of stillborn piglets and mummies than those with Patterns 2-MH and 4-MM (P < 0.01). CONCLUSIONS: This study indicates the importance of reaching Pattern 1-HH by rapidly increasing feed intake during early lactation and high feed intake during late lactation, which is associated with high weaning performance and subsequent reproductive performance of the sows. Also, the current study suggests that Pattern 1-HH is linked to good farrowing with a low percentage of stillborn piglets and mummies. Finally, it is critical for producers to timely identify a problem of sows' eating behavior and to make a prompt decision to intervene.

Diseases caused by Escherichia coli are recognized as major problems in the swine industry, one of them being edema disease (ED). Importantly, the current decrease in antibiotic use may cause difficulties in controlling the disorders caused by E. coli. Therefore, this study assessed the efficacy of a commercial vaccine against ED in nursery pigs from a farm with previous history of ED. A total of 1344 pigs were monitored; half of them were randomly assigned to a vaccinated group (VG) and the other half to a non-vaccinated group (NVG). The vaccine was administered at 7 days of age. Animals received a pre-starter feed with 2500 ppm of zinc oxide (ZnO) for 2 weeks and a starter feed without ZnO for another 3 weeks. Pen-group weights were recorded at 28 (weaning), 42 (end of pre-starter phase), and 63 days of life (end of nursery phase). Death/culling rates, average daily gain (ADG), and average daily feed intake (ADFI) were calculated for each group at each phase. The overall relative risk of dying/being culled for a pig in the NVG was 5 times higher than that of the VG group but increased to 12 times higher during the starter period. ADG and ADFI were also significantly higher in the VG group for that period. Vaccination against ED significantly reduced pig losses and improved ADG and ADFI, particularly when ZnO was not used.

Evidence-Based Psychological Treatments for Adults: A Selective Review. BACKGROUND: Psychological treatments have shown their efficacy, effectiveness, and efficiency in dealing with mental disorders. However, considering the scientific knowledge generated in recent years, in the Spanish context, there are no updating studies about empirically supported psychological treatments. The main goal was to carry out a selective review of the main empirically supported psychological treatments for mental disorders in adults. METHOD: Levels of evidence and degrees of recommendation were collected based on the criteria proposed by the Spanish National Health System (Clinical Practice Guidelines) for different psychological disorders. RESULTS: The results indicate that psychological treatments have empirical support for the approach to a wide range of psychological disorders. These levels of empirical evidence gathered range from low to high depending on the psychological disorder analysed. The review indicates the existence of certain fields of intervention that need further investigation. CONCLUSIONS: Based on this selective review, psychology professionals will be able to have rigorous, up-to-date information that allows them to make informed decisions when implementing empirically based psychotherapeutic procedures based on the characteristics of the people who require help.

BACKGROUND: Primiparous sows (PP) have higher nutrient requirements, fewer piglets born with lower birth weight and growth performance than multiparous sows (MP). The aim of the current study was to investigate the effect of parity of sow (PP or MP) on the growth performance and humoral immune response of piglets. A total of 10 PP and 10 MP (3rd to 5th parity) sows were used. There were 4 treatments in a 2 × 2 factorial arrangement, with piglets from PP sows suckled by PP or MP sows, and piglets from MP sows suckled by PP or MP sows. Average daily gain (ADG) of piglets during the lactation period, and ADG, average daily feed intake (ADFI) and gain:feed ratio (G:F) from weaning to 144 days of age were controlled, and concentrations of immunoglobulins G (IgG) and major acute phase protein (Pig-MAP) were measured as markers of humoral immune response throughout the study. RESULTS: Total ADG was higher in piglets born from MP than in those born from PP (669 vs. 605 g/day; standard error of the mean (SEM) = 15.5, n = 5; P = 0.001) and in piglets suckled by MP than in piglets suckled by PP (655 vs. 620 g/day; SEM = 15.5, n = 5, P = 0.037). Total ADFI was higher for pigs born from MP than for those born from PP (1592 vs. 1438 g/d, SEM = 42.2, n = 5, P < 0.001). Total G:F tended to be higher for pigs suckled by MP than for those suckled by PP (0.43 vs. 0.41, SEM = 0.006, n = 5, P = 0.076). At weaning, IgG serum concentration was higher (30.0 vs. 17.8 mg/mL, SEM = 4.98, n = 15, P = 0.013) in pigs suckled by MP than in piglets suckled by PP. However, IgG concentrations were higher for pigs born from PP than for pigs born from MP on days 116 (P < 0.001) and 144 (P = 0.088). Pig-MAP tended to be lower in pigs suckled by MP than in pigs suckled by PP on days 40 and 60 of age (P < 0.10). CONCLUSIONS: The research indicates that the growth performance and humoral immune response of the offspring of PP is improved by cross-fostering with MP. These results open the possibility of an interesting strategy for improving the growth of litters from PP, that is easier to apply than current programs based on parity segregation, which implies a separate building site to house gilts, first parity sows and their offspring.

The objective of this study was to assess the effects of a probiotic strain Clostridium butyricumMIYAIRI 588 (CBM588) on broiler and weaned piglet health and zootechnical performance. Five field studies were carried out in broilers and five in weaned piglets under European feed additive guidelines. Each study followed a randomized blocked design with two treatments: Control (basal diet) and CBM588 supplemented groups. The zootechnical performance parameters selected were body weight, daily gain, feed intake and feed efficiency (feed:gain). Broilers fed diets with CBM588 gained significantly more weight (+2%, p < .001) and exhibited significantly better feed efficiency (-1.6%, p < .001) in comparison with Controls. Similarly, analysis of pooled data of weaned piglet trials showed that CBM588-fed piglets were significantly heavier than Controls (+2.6%, p = .014), exhibited significantly higher mean daily gain (+4.7%; p = .004), and significantly improved feed efficiency (-4.2%, p = .001). In addition to the zootechnical efficacy studies, the preventive effect of CBM588 on necrotic enteritis (NE) was assessed in a natural challenge model in broilers where CBM588 reduced the incidence and severity of NE lesions. These data indicate the potential of CBM588 to improve broiler and weaned piglet zootechnical performance, and to make a positive contribution to animal health.

BACKGROUND: Surgical castration is still practiced in many EU countries to avoid undesirable aggressive behavior and boar taint in male pigs. However, evidence shows that castration is painful and has a detrimental influence on pig health. This study investigated the clinical and productive effects of surgical castration in the suckling period. A total of 3696 male pigs, 3 to 6 days old, comprising of 721 litters from two different farms were included in the study. Within each litter, half of the males were kept as intact males (IM) and half were surgically castrated (CM). Surgical castration was conducted by a trained farmer. Average daily gain (ADG), body weight at weaning (BWW), percentage of pre-weaning mortality (PWM) and antibiotic usage were measured. Pig major acute phase protein (PigMAP) serum concentrations were analyzed prior to castration, and on days 1 and 10 after castration. Productive performance data were analyzed using a linear mixed model. Mortality and percentage of pigs treated with antibiotics were analyzed using the Fisher's exact test. RESULTS: No overall differences in BWW and ADG were observed between the two groups. However, differences were observed when the same effects were analyzed in the 25% lightest, 50% medium and 25% heaviest pigs at birth. PWM was higher in CM than in IM groups (6.3% vs 3.6%; p < 0.001), especially in the light (12.2% vs 6.2%; p = 0.02) and in the medium (5.5% vs 2.7%; p = 0.04) weight groups. In the heaviest pigs group PWM was not affected by castration, but IM tended to show higher ADG (p = 0.06) and showed higher BWW (8.0 kg vs 7.8 kg; p = 0.05) than CM. There were no differences in percentage of pigs treated with antibiotics between the two groups (5.8% vs 5.8%; p = 0.98) in this study. Furthermore, PigMAP was increased in CM the day after castration (0.944 mg/ml vs 0.847 mg/ml; p = 0.025), but there was no difference between CM and IM groups at day 10. CONCLUSIONS: Surgical castration has a negative impact on production in the suckling period because it causes an increase in PWM, especially in pigs in the three lower quartiles for body weight, and negatively affects the BWW in pigs born in the highest quartile for body weight.

The serum concentration of acute phase proteins (APPs) increases in the presence of disease or stress, which makes APPs notable parameters for the global assessment of animal health and welfare. A rapid, immunochromatographic test (ICT) for the detection of elevated levels of pig Major Acute-phase Protein (pig-MAP), one of the main APPs in pigs, was evaluated in more than 1400 pig serum samples obtained from commercial farms. The ICT showed a good performance with a relative sensitivity (Sn) and specificity (Sp) of 94 and 97%, respectively, for a threshold of 1.5mg/mL (comparison with ELISA). Differences in the pig-MAP levels and the number of positive samples with the ICT were observed within the season of sampling, farms, and age groups at one farm, according to the presence of disease or lesions. The ICT was also evaluated in blood samples obtained at slaughter in association with the carcase inspection. The results from this study indicate that the ICT may be used for the evaluation of groups of pigs, after analysing one sub-sample of these pigs, and might be a useful tool in routine health and welfare monitoring programmes aimed to improve the quality of pig production.

We studied the seasonal fluctuation of soil respiration (R(S)), and its root-dependent (R(R)) and basal (R(B)) components, in a Vitis vinifera (Chardonnay) vineyard. The R(S) components were estimated through independent field methods (y-intercept and trenching) and modeled on the basis of a Q(10) response to soil temperature, and fine and coarse root respiration coefficients. The effect of assimilate availability on R(R) was assessed through a trunk girdling treatment. The apparent Q(10) for R(R) was twice that of R(B) (3.5 vs 1.6) and increased linearly with increasing vine root biomass. The fastest R(R) of fine roots was during rapid fruit growth and the fastest R(R) of coarse roots was immediately following fruit development. R(S) was estimated at 32.6 kg ha(-1) d(-1) (69% as a result of R(R) ) for the hottest month and at 7.6 kg ha(-1) d(-1) (18% as a result of R(R)) during winter dormancy. Annual R(S) was low compared with other natural and cultivated ecosystems: 5.4 Mg ha(-1) (46% as a result of R(R)). Our estimates of annual vineyard R(S) are the first for any horticultural crop and suggest that the assumption that they are similar to those of annual crops or forest trees might lead to an overestimation.

Pig-MAP (Major Acute-phase Protein) and haptoglobin concentrations were determined in pigs from commercial farms, and reference intervals obtained for different productive stages. Pig-MAP serum concentrations were lower in sows than in adult boars (mean values 0.81 vs. 1.23 mg/mL) and the opposite was observed for haptoglobin (1.47 vs. 0.94 mg/mL). No differences were found between parities, except for a minor decrease in haptoglobin concentration in the 4th parity. A linear correlation between pig-MAP and haptoglobin concentration was observed. In the period 4-12 weeks of life, pig-MAP mean concentrations were around 1mg/mL, being lower in the finishing period (0.7-0.8 mg/mL). Haptoglobin concentrations increased with time, from around 0.6 mg/mL at 4 weeks of age to 1.4 mg/mL at 12 weeks. Mean values of around 0.9 mg/mL were observed in the finishing period. A wider distribution of values was observed for haptoglobin than for pig-MAP concentrations. Differences between herds were observed, with the highest values obtained in a herd with signs of respiratory disease.

Desde la Teoría Racional Emotiva de Albert Ellis se distingue entres ansiedad primaria y secundaria, habiéndose, en pocos casos, ignorado este hecho en los tratamientos psicológicos (como la exposición, donde se pretende la habituación a la ansiedad primaria). Este artículo pretende analizar la diferencia entre los dos tipos de ansiedad en un paciente, tratado en un dispositivo público de salud mental, con trastorno de pánico y las consecuencias de esta diferenciación en la utilización de técnicas terapéuticas


Albert Ellis´ rational emotive theory differs between primary and secondary anxiety but this differentiation has been ignored in the psychological treatments (for example in the exposition, which try the habituation to the primary anxiety). This paper tries to analyze the difference between both types of anxiety in a patient, attended in a public device on mental health, diagnosed of panic disorder and the consequences that stem from this differentiation in the employment of therapeutics techniques

The acute phase protein (APP) response was evaluated after prolonged transportation of pigs under commercial conditions. Elevated serum APP concentrations were observed in two groups of boars immediately after their arrival at a destination farm compared with within-animal control samples obtained one month later. The effect was more pronounced in the first group of pigs conveyed under average transport conditions (Transport 1, 24 h), although the second group was transported for a longer time period (Transport 2, 48 h) but in superior transport conditions. In a second trial, pigs were sampled before transport, on arrival at an abattoir (following 12 h transport), and at the slaughter-line (after 6 h lairage). Significant increases in major acute phase protein (Pig-MAP), haptoglobin, serum amyloid A, C-reactive protein, and a decrease in apolipoprotein A-I, were observed at slaughter. The results demonstrate that shipment of pigs by road can result in an APP response that is probably related to the stress of transport.

The objective of this study was to determine the serum concentration levels of selected acute phase proteins (APP), haptoglobin (HPT) and pig-major acute phase protein (pig-MAP), in postweaning multisystemic wasting syndrome (PMWS) affected pigs and PCV2-subclinically infected pigs. In a first study, a group of 15 eight-week-old conventional pigs from a PMWS affected farm were bled and a complete necropsy, histopathology and in situ hybridisation to detect PCV2 were performed. Based on the results, pigs were classified as suffering from PMWS (n = 10) or healthy animals (n = 5). In a second study, a group of 45 pigs from another PMWS affected farm were selected and bled at 3, 7, 12 and 28 weeks of age. The assessment of PCV2 infection status in these pigs was retrospectively done by PCV2 PCR in serum samples. Selected APP were measured in the serum of all studied pigs by means of radial immunodiffusion. Mean HPT and pig-MAP levels were significantly increased (p = 0.004 and p = 0.0006 respectively) in PMWS-affected pigs when compared to levels found in healthy pigs (2.52 +/- 0.88 mg/mL vs. 1.06 +/- 0.73 mg/mL for HPT and 3.81 +/- 1.53 mg/mL vs. 0.76 +/- 0.34 mg/mL for pig-MAP). In the second study, no significant difference in mean HPT and pig-MAP values were observed between PCV2 PCR positive and negative pigs of any age. However, both APP increased significantly with age in PCV2 PCR negative pigs. Altogether, the present results suggest that APP levels are significantly increased in pigs that develop PMWS, but not in animals with a PCV2 subclinical infection.

Male growing pigs were fed a diet containing 250 g/kg of native corn starch (CS; 26% amylose, 74% amylopectin) or 250 g/kg of raw potato starch (RPS), as examples of digestible starch and resistant starch (Type II), respectively. Whole-tract digestibilities of organic matter, crude protein and starch were greater in pigs fed CS than in those fed RPS through at least d 23 of the study. However, the values progressively increased in the RPS-fed pigs up to d 38, at which time the groups did not differ in organic matter and starch digestibility. The digestive tract and colonic digesta were heavier and colon length longer in pigs fed the RPS diet. Digestibility of starch in the ileum on d 38 was significantly lower in RPS-fed pigs, but rose from ileum to rectum; most starch was extensively fermented in the cecum and proximal colon. Purine base (PB) and short-chain fatty acid (SCFA) concentrations in feces initially increased and then decreased beginning on d 4 for PB and on d 21 for SCFA. PB concentration in feces was greater in pigs fed RPS than in those fed CS. In the large bowel digesta, PB and SCFA concentrations increased from the ileum to the cecum and proximal colon and then fell in the distal colon. Pigs fed the RPS diet had a higher PB concentration in the middle colonic digesta and a greater SCFA concentration in the proximal colonic digesta than the CS-fed group. Adaptation of growing pigs to supplementary RPS required 5 wk, as reflected by whole-tract digestibility and PB and SCFA fecal excretion data.

Por lo expuesto en todo el curso de los capítulos anteriores podemos concluir en lo siguiente: 1° Que el índice de parasitosis en el grupo de 4 a 6 años es del 76 por ciento. 2°. Que los cuatro parácitos más frecuentes fueron, Entamoeba Histolítica (38 por ciento), Ascaria Lumbricoides (17 por ciento), Blastocistis Hominis (12 por ciento) y Tenia Hymenolepis Nana (8 por ciento ). 3. Que los casos de mono parasitosis fueron 55, con dos parásitos 18, con tres parásitos 3 y 24 placas negativas. 4. Que el sexo femenino es el más afectado en los siguientes parásitos: Entamoeba Histolítica, Hymenolepis Nana, Enteromonas Hominis, Trichuris Trichura, Dientamoeba, Monilias, Hongos. 5° Que en el sexo masculino se encuentran equilibrados o en mayor número el resto de los parásitos que no figuran en la cuarta conclusión. 6°. Que la mayor incidencia de parasitosis se encuentra entre los 6 y 8 años de edad. 7°. Si bien se encontró el 76 por ciento de incidencia parasitaria, de haberse utilizado un método más minucioso, como el de enriquecimiento y cultivo o la repetición de las placas negativas, cimiento y cultivo o la repetición de las placas negativas, este índice aumentaria porcentualmente. 8°. Que en el presente tabajo no se ha encontrado ni un solo caso de Esterobius Vermicularis, debido seguramente a medios indecuados de pesquisa para este parásito. 9°. Por último debemos recomendar la necesidad de vigilancia por organismos sanitarios y la prestación de medios profilácticos, diagnóstico y de tratamiento para la solución tan grave problema
